The laboratory forms part of the newly formed Oxford Centre for Translational Myeloma Research (http://oxford-myeloma.org.uk/). The Centre is a hub to collaborate with national and international teams to perform research into myeloma and develop novel therapeutic interventions. This post will work closely with pharmaceutical partners and apply single cell techniques to understand development of drug resistance and identification of novel drug targets and mechanisms. Unifying the diverse research areas in the team is the extensive use and development of novel Next Generation Sequencing (NGS), flow or mass cytometry (Cytof) and spatial multiplexed imaging based methods. The individual holding this position will employ a combination of tissue culture and molecular biology techniques,flow and mass cytometry, genetic manipulation (CRISPR/CAS; sh/siRNA, and compound screening. Cell based assays for mechasnism of action of drugs would be part of the responsibility. This role will support the research of Prof Thakurta in multiple projects. Key responsibilities • • • • • • • • • • • • • • • • • Cell culture and biological functional assay development Cell biology and biochemical assays for understanding drug or disease mechanisms. Flow and mass cytometry (Cytof) NGS based sequencing of cell lines Proteomics experiments to characterize cell and immune cells Generation of resistant preclinical models Working with Organoid models of Myeloma PBMC or BMNC Cell sorting and cell type specific PD and phenotypic assays Co culture based cell killing and other assays for drug combinations and drug MoA Cell culture-based manipulation - eg cloning, stable cell line generation, CRISPR/CAS compound screening using semi-automated, low-medium throughput approaches. General laboratory maintenance, e.g. maintain stocks and order laboratory supplies and consumables Teaching experimental techniques to new laboratory members and providing supervision of new lab members within the laboratory Liaising with direct supervisor, and other members of group, regarding experiments and general laboratory procedures To participate in networks and other working groups Written communication of experimental reports and data deposition in electronic laboratory notebook and database Undertake other duties as may be required from time to time that are commensurate with the grade and responsibilities of the post.
