Our research group is focused on studying the UbiquitinProteasome System, a critical regulator of the fate of most cellular proteins. We are interested in discovering aberrant function of the UPS in the context of cancer immunology, and how this can be exploited for the development of novel therapeutics. The main responsibility of this post is to support the laboratory in the characterization of cancer immunomodulatory target genes by different proteomic, biochemical, and immunological techniques. In addition, you will support industry-related activities, including small molecule inhibitor characterisation by studying their activity and selectivity profile, in vitro and in living cells, using activity-based probes and a number of proteomic approaches. In addition, the effects (viability, migration, metabolism, immunogenicity…) of these genes and drugs in cancer cell models will be evaluated in combination with genetic impairment of the selected target gene using knockdown/CRISPR knockout experiments. Responsibilities You will: • • • • • • • • • Perform experiments examining deubiquitylase (DUB) activity profiles using molecular active site probes and small molecule inhibitors. Perform research work with human and mammalian cell lines (tissue culture), inlcuding the generation and maintenance of knockout and knockdown cell llines using CRISPR/siRNA. Prepare mass spectrometry (MS) samples from a variety of biological sources (i.e. patient tissue biopsies, cell cultures, blood, peripheral blood mononuclear cells (PBMCs), etc). Perform functional validation assays including FACS, ELISA, and Incucyte.. Assist with complex proteomics and mass spectrometry data analysis. Be responsible with others for the biological safety of the laboratory. Perform laboratory tests in order to produce reliable and precise data to support scientific investigations. Keep detailed record of all activities and to consolidate data from various projects within the group, interpret the results and to present them to other team members, including external collaborators.
